Emphasis has been placed, in particular, on the rationale for the development of second-generation calcium antagonists, and the advantages associated with their use. An attempt has been made to describe the conditions under which the second generation of antagonists offer specific advantages over the first generation, and to explain the basis of the tissue selectivity exhibited by some of these newer drugs. To this end, explanations are presented of the recent advances which have been made in unravelling the molecular architecture of the calcium channel and itsassociated calcium antagonist binding sites. The book has been prepared in such a way as to be readable, and does not aim to cover the entire field; rather specific second-generation calcium antagonists are described as examples of this new direction of development in the overall field ofcalcium antagonism.
